She made her film debut the same year with My First Romance. Her career breakthrough came with the 2007 film One More Chance, which became a major box office hit and earned her FAMAS and Luna Awards for Best Actress. She followed this with the hit sitcom I Love Betty La Fea (2008–2009), a Filipino adaptation of the Colombian telenovela Yo Soy Betty, la Fea.

Through the 2010s, Alonzo starred in a string of commercially successful films, including The Mistress (2012), Four Sisters and a Wedding (2013), She’s the One (2013), A Second Chance (2015) — which became one of the highest-grossing Filipino films of all time — and The Love Affair (2015).

In 2021, after nearly two decades with ABS-CBN, she transferred to GMA Network. She launched her YouTube channel, #ByBea, in 2023, and starred in the GMA series Love Before Sunrise (2023) and Widow’s War (2024–2025) alongside Carla Abellana.
